Description

GENERAL SUMMARY/ OVERVIEW STATEMENT:

The Research Technologist in the Bei laboratory/Department of Surgery located at the 2nd floor of Shriners Boston is a full-time position under the direct supervision of the PI, Dr. Bei. Our research focuses on understanding the mechanisms that regulate craniofacial formation, homeostasis under normal and pathological conditions and how skin repairs and regenerates after wounding. We focus extensively on the role of transcription factors and post-transcriptional regulation of gene expression and cell fate. This Technologist independently carries out a broad range of technically advanced research activities and procedures; evaluates conclusions and has considerable latitude to modify or devise methods and techniques as necessary to achieve desired results. Experience in working with genetically modified animal models, molecular biology methods, DNA/RNA sequence analysis, histology, in situ hybridization, Immunohistochemistry, and cell culture is required

PRINCIPAL DUTIES AND RESPONSIBILITIES:

  • Directs and carries out experiments independently on projects of the lab under the advice of the PI and postdoctoral fellows.
  • Trains and provides ongoing guidance for technicians and students in the use of laboratory equipment.
  • Generates and maintains a book of laboratory protocols for the lab use.
  • Helps maintaining the laboratory environment and manage the reagents used in experiments.
  • Keeping lab inventories (cell lines, antibodies, mouse mutant lines), updating lab archives in collaboration with the PI and postdocs
  • Maintaining all Bei Lab equipment and is responsible for the annual inspection of lab equipment, ordering, and keeping general Bei lab environment.
  • Independently performs molecular biology experiments, including: molecular cloning, isolation of DNA and RNA from cells, or tissue, evaluation of RNA quality using the Bioanalyzer; quantification of DNA qualify/quantity using the Nanodrop; primer design; RT-qPCR amplification of genes; preparation of PCR samples for NGS sequencing; analysis of sequence for mutations and assessment of functionality using various databases/resources; cell culture; cell transfections and transductions; lentivirus vector preparation; fluorescent microscopy; protein quantification; restriction digests of DNA; ISH, immunocytochemistry; western blots.
  • Work with animal models (maintenance and genotyping), handling discarded mouse embryonic and adult tissue samples, and keeping detailed records.
  • Must have good experiences of animal studies in compliance to all the guidelines of the use of animals in biomedical research.
  • Develops research methodologies within the parameters of experimental protocols and research objectives and in collaboration with the PI and postdoctoral fellows.
  • Responsible for oversight and coordination of scheduling of experimental procedures.
  • Orients and trains new research technicians as needed.
  • Responsible for troubleshooting problems and instructing others in highly specialized techniques.
  • Synthesizes results in collaboration with the PI and postdoctoral fellows.
  • May perform data analysis using statistical techniques as needed.
  • Composes and may present the method sections of research reports and manuscripts as needed.
  • Expertise in most computer programs used by the lab.
  • Provides administrational support for the lab if needed.
  • Participates in all lab meetings.

SKILLS & COMPETENCIES REQUIRED:

  • Ability to make independent, effective decisions and troubleshoot instruments.
  • Demonstrated competence in advanced research techniques and methodologies.
  • Strong organizational and communication skills.
  • Ability to work in a group environment and be of assistance to a number of research projects.
  • Demonstrated analytical skills to problem-solve effectively.
  • Functions with speed, precision, and an understanding of the theory behind tests and procedures.
