10010 N Torrey Pines Rd, La Jolla, CA 92037The lab manager position is dual in scope: as a bench scientist supporting studies in the lab and as a laboratory manager. Alan Saghatelian's laboratory uses next-generation nucleotide sequencing and mass spectrometry to identify novel microproteins and define their functions in cells and in mice. This position places significant emphasis on the bench scientist role. The candidate will spend most of their time using cell and molecular biology techniques to support peptide discovery projects from hit identification to validation and functional studies. Lab manager duties will include ordering supplies, managing the database of lab materials, and assisting the PI with budgeting. The candidate will also supervise and train junior technicians, students, and postdoctoral fellows in molecular and cell biology techniques.Who We AreThe Salk Institute is an internationally renowned research institution that values diversity, equity, and inclusion. From visionary leaders to dedicated administrators and brilliant faculty members, the Institute is united by a shared passion for scientific exploration and innovation.What Your Key Responsibilities Will Be1) Research Support (70%)The candidate will use a variety of molecular biology techniques, including designing DNA primers for PCR, ordering DNA constructs and plasmids, and confirming their fidelity using restriction enzymes and gel electrophoresis. The candidate should be familiar with Gateway and Gibson cloning methods and software such as SnapGene.Responsible for routinely growing cells in culture, transfecting cells with plasmids to express proteins of interest, and establishing stable cell lines. Studies may require measuring cell secretion products using commercial ELISAs.Use cell surface receptor signaling assays (time-resolved fluorescence and luciferase reporter) to determine the structural requirements of microproteins for binding and activation.Review relevant literature to solve problems and modify and adapt techniques to implement new protocols for lab studies.Use plate-based fluorescent and luminescent activity assays for in-vitro pharmacology of various targets.Develop and execute experiments involving PCR, sequencing, and other DNA-based techniques.Prepare cells and tissues for RNAseq and Riboseq analysis and maintain the workflow for submission to Institute core facilities for sequencing.Assist post-doctoral fellows and provide technical expertise, including carrying out CRISPR screens by preparing lentiviral libraries and cell selection or FACS.2) Laboratory Management (20%)Manage the ordering of reagents and supplies for lab projects using purchasing software. Approve all orders and, together with the PI, manage the funding to be used.Manage freezer inventory and databases of plasmids and reagents in the lab in electronic format.Oversee general lab maintenance and safety. Inform new personnel of required safety procedures and ensure safety updates are disseminated to laboratory personnel.With assistance from postdoctoral fellows, train and oversee junior technicians and undergraduate students in molecular and cell biology techniques.Communicate and interact with Salk departments (HR, accounting, environmental health, materials transfer) and external vendors to conduct necessary lab business.3) Other Duties as Assigned (10%)Analyze data, prepare graphics using standard programs and present in group meetings.Ensure quality and integrity of data collected and maintain data in electronic form.Assist PIs and postdocs with grant and report writing and study-related publications.Supervisory Responsibilities:This job has no supervisory responsibilities.What we RequireBachelor's degree in biology or related field.Minimum of 5 years of advanced experience in molecular and cell biology, including design of DNA constructs and sequencing and in-vitro cell-based assays with ELISAs or CRE-luciferase or time-resolved fluorescence readouts.Minimum 3 years of experience in a supervisory/managerial role within a laboratory setting.Strong collaborative and written and verbal communication skills.Preferred:Experience with receptor signaling assays. Experience with the preparation of samples for riboseq or RNAseq.What We Can OfferThe expected pay range for this position is $82,000 to $102,750 annually.
