The successful candidate will provide key mechanistic, preclinical, and translational immunology support for Cue Biopharma’s growing pipeline of therapeutic candidates for the treatment of cancer and autoimmune disease. The successful candidate will also contribute to understanding the breadth of disease-relevant T-cell receptor (TCR) repertoires as well as to generating and interrogating TCR-transduced human cell lines and primary human T cells.

Responsibilities

Plan, execute and analyze

in vitro / ex vivo

studies using primary human immune cells (from healthy donors and patients).

Perform immunophenotyping using multicolor flow cytometry, FACS, and cellular immunology assays.

Isolation, viral transduction and culture of primary human/non-human PBMCs and immune cell subsets.

Expansion, selection and long-term maintenance of T cell clones/lines.

Contribute to all aspects of generating TCR lentiviral/retroviral vectors including design, construction and testing.

Conduct experiments and accurately document and effectively communicate data with project team, colleagues and manager.

Ph.D. in Immunology or a relevant field with 0-5 years of related laboratory experience, M.S. in a relevant field with 2-7+ years of related laboratory experience, or B.S. in a relevant field with 5-9+ years of related laboratory experience.

Hands-on experience executing and documenting

in vitro

biology studies of primary human immune cells is required.

Multicolor flow cytometry-based immunophenotyping, FACS, cellular immunology

in vitro

assays (i.e., cytokine analysis, cytotoxicity, and proliferation) is highly desirable.

Hands-on experience analyzing antigen-specific T cells is a plus.

Direct experience with molecular biology, lentiviral/retroviral vectors, synthetic biology and assay development is highly desirable.

Hands-on experience generating stably transduced T cell lines and primary T cells; knowledge of TCR V(D)J recombination is a plus but not required.

Experience with B cells, granulocytes, and/or therapeutic targeting in the context of autoimmunity is a plus.
