This role is considered On-Site.OverviewThe Delivery Sciences team is seeking a Scientist to contribute to our ligand discovery efforts for expanding RNAi to extrahepatic tissues. This position will focus on identifying, characterizing, and optimizing peptide and protein ligands for targeted delivery of RNAi therapeutics. This person will work closely with multiple groups across the research organization. The position is onsite and will be primarily located at 675 W Kendall St Cambridge MA.Key ResponsibilitiesDevelop and optimize binders (peptide, VHH, mAb etc.) targeting conformationally diverse membrane proteins using molecular evolution techniques and affinity maturation strategies (e.g. phage display, yeast display, etc.)Characterize binders using a variety of biochemical techniques (ELISA, BLI/SPR, epitope binning) and in vitro assays (fluorescence microscopy, flow cytometry)Work collaboratively with multiple groups across the organization to evaluate ligands for tissue-specific delivery of RNAi

and enable them for developmentDesign and execute well controlled experiments, analyze data, and present findings to various groups across the research organizationQualificationsPhD in biochemistry, biology, or a related field with 0-3 years of work experience in an academic or industry settingExpertise in peptide, scFv, and/or VHH library design, construction, and validationExpertise with identifying and optimizing binders targeting a broad range of proteins with high affinity using phage and/or yeast display and affinity maturationExperience with plasmid design for recombinant protein expression in bacterial and mammalian hostsExperience with preparative scale protein purification (IMAC, affinity, IEX, SEC) and standard recombinant protein characterization techniques (UV-vis, SDS-PAGE, Western blotting, ELISA)Experience with kinetic characterization of ligand/receptor interactions by BLI and/or SPRStrong interpersonal skills and excellent communication skills (written and oral) are requiredAbout Alnylam:
